How do I remove a follower of mine on twitter and or block them?

You can block a twitter follower by going onto his profile, clicking the right symbol next to message and lists. Then click on <username> block and you're good to go.

Just go to his/her twitter profile and hit the Block button. After that, he/she can not visit your profile and disabled in reading your tweets.
