I read over the site, and can't really get a grasp on what it's for, and exactly what it does.
What are the practical uses for it? What makes it worth signing up for?

Basically it keeps you up to date with what your fave celebs/friends are doing . You follow certain people then on your home page their statuses appear there.

same with me....i have also signed up on twitter but never loged in again.....wot my friend told me that it is a socialising site where u follow famous personalities and read their blogs

Twitter is a microblogging site that helps you broadcast short statuses or messages to a community of followers.

It is very handy to diffuse news, report on live events, analyse search and internet trends...etc
Think about your Facebook status update being broadcasted to a wide audience and throughout the web.

It is worth using if you have regular news or updates you want your follower's or customers to know about.
It is also very handy as a mobile way of communicating.
