Well, you must find any Maria's message and reply to it (there is button near every message). But actually Twitter is not place for messaging to each other or chating. You can do it on MySpace or Facebook. In twitter you better only to post something.. (140 symbols).
It is like a social network for "statuses". It is like a blog, but very small.. If you "follow" somebody you will see their messages on your homepage. And if you want, you can reply to them something....

Example: you may post something like "I'm eating soup now"
or "I hate my life" :-))) or anything you want... and you will see posted messages of people whom you are following. That's all....

twitter is pretty much a more improved version of the status updates feature of facebook and that's it. to communicate with maria for example, if her username is 'maria' you need to enter an update starting with @maria and type 'how are you' this goes to her in her replies tab but visible to everyone, to talk to her directly, you need to use the direct messages feature which is private.

if someone wishes to see your updates in their 'timeline' they follow you, if you follow someone you see their updates in your timeline, the timeline is what you see when you login to your twitter home page. did i miss anything?
