When you're on the page with the video on, click: Edit video. Then go to Privacy and select: Customize. It then comes up with a page that says Custom Privacy. Type in your sister's name where it says: Hide this from these people. And then save!

Do like Didy said, but remember: she can ask her friend (who is also your friend) to let her/him see it.
So if you really don't wan't her to see it, better don't post it at all.
