You have to start by following alot of people
then those people may or may not follow you back
so whenever someone follows you follow them back

You should try automating your twitter. I have this software and it's better than any software I used because I can auto tweet, auto message and do a whole lot more. Also, I can search for who I want to follow me five different ways such as by subject, location (which is great for realtors and other business owners) and more. You can get a lot of followers this way. There is nothing but targeted followers who are interested in your subject matter.
Also, you want to have interesting content on your website in order to keep your followers.

The best way to get followers is to basically search for users that share the same interests as you do. Once you have a list you can start following them and between 30 & 60 percent will follow you back. I use software from http://www.tweetwhistle.com to do this. You can do it manually but spend a few bucks and let the software do it for you...It's faster and easier.
Another thing to note is once you reach 2000 followers you can only follow 10% more then the number following you but not to worry the software I mentioned above allows you to unfollow any number of people so you can start the entire process over again.

Write a bio that explains something about yourself, add a homepage etc. to show people that you are really a real person.
Follow much people. Tweet alot. Retweet, @mention, add #tags and use Trending Topics as a way to be noticed.
