I posted on twitter this:
''Would you hold it against me''
And this person/my friend replied:
''Why yes...yes I would.''
What does he means with that?

Haha, sounds like he's playing around. I could see my friends saying that just to be like "I would" they mean it in a playful way. It's nothing to worry about. "Would you hold it against me?" is kind of a funny post, it's like a play on the song, so they were just having fun, and maybe being flirty.
