While you're signed in on twitter.com, go to 'find people' on the top right of the page. Then type whatever you're looking for into the search engine. I'm not sure what kind of followers you want, but type a keyword such as 'guitar' or 'hockey' into the box and you'll find people targeted to your needs/wants. Following people usually leads to them following you back.. so that is the easiest way of getting valuable/targeted twitter followers, mainly because you get to choose who you want..

I find a great way to get Twitter followers is by using a Twitter train. You goto the site, sign up, then follow people. Then you get added to the train, and the new people follow you. Give this one a try: http://targetfollowers.com
There is also a software called Phoenix Twitter Desktop that allows you to follow users in bulk by keyword, or follow another user's followers (that might be in the same niche). It works great, but I warn you, don't follow or unfollow more than 500 a day. You can find yourself locked down by Twitter, or even being removed.
