Like, i have how ever many followers on twitter now, but if i lock my tweets, will they have to send me a request again or will it not matter and they'll already be unlocked for them?

If you lock your tweets, only people who are following you will be able to see them and people that wants to follow you but hasn't yet will not be able to see them.
