I want to use twitter to promote my website how should I go about this?

There are many ways to promote your website. Twitter is one of the bookmarking site. Here's what you can do:
Here are the list of ways that you can do:
1. Post blogs on any social media networks about your website.
2. Submit articles to any article sites.
3. Social bookmarking. Bookmark your site (@ socialmarker.com)
4. Submit your website to search engine directory.
5.Use keywords liberally on your site as long as they are relevant to your site's content and topics.
6.Reciprocal linking

A lot of different ways to go about this.
A really effective way for you to go about doing this is by going to http://search.twitter.com, typing in "need cell phone" or "need a new cell phone" and look for recent tweets pertaining to people needing a new mobile phone. A great way to target people directly.

Create keywords cloud , and compare with others talking about similar niche, best tool for it is Contacted http://www.contacted.org . Use your website keywords with contacted

Don't promote your website.
Twitter is a social networking application not an ad network. Use Twitter to build up relationships, contacts and 'friends' and just have your link in the URL box. People will click it, once they get to know and like you as a person.
Social and business networking is about selling the benefits of the individual, the websites will follow much quicker that way and you'll develop followers naturally.
Straight advertising will turn people off and they won't bother clicking.
