Log in to Twitter via the Twitter Home Page, or via a Twitter API application such as Twhirl. You can also update Twitter via Facebook and several other applications that use the Twitter API.

Enter your message in the box with the label "What are you doing?" Keep in mind that your tweet can be no longer than 140 characters at the most. This limitation exists because Twitter uses the SMS network to send notifications via cell phone to those who desire it.

Click "Update" to send the Tweet to everyone on Twitter.

Type "d USERNAME" before your tweet to send a direct message to a particular user. Only that user will see the message.

Put "@USERNAME" at the beginning of your tweet to send a reply to a particular user. That user, your followers and that user's followers will all be able to see the tweet, but it won't appear on the public timeline.
