If I am following someone but they aren't following me and they ask a question. If I press respond and send a "@twittername" tweet, will they see it?

They can see if they go to their @replies because your tweet won't show up on their homepage.

Yes, @twittername will let them see it. Once they follow you back, you can send them a direct/private message.
