I know that they are for famous or well known people, but how exactly can they be sure that it is the right person? Does the persons manager have to contact Twitter or something?

A verified twitter account means that they have verified that you are human by requesting a legitimate phone number be entered. They send you a code that you then type into the website and then become verified.
