Quite simple, an easy way is to follow lots of other people/groups etc. When they see someone following them, most people tend to follow you then.

the most important thing is to create interesting content - people follow you if you have some insight to share and interesting helpful tips and not just "I just ate vanilla icream"
of course you can use software such as TweetUnlimited.com that will mass message people
and you will sure get attention but it might be short lived
the count of the followers size does nothing say about you - if you provide interesting things and if they are specific to one topic you could be known on Twitter to be that "topic" person and soon find thousands of people following you like a celebrity
