I've seen lots of them at the end of tweets and there are also some trending ones. I'm new and I'm wondering how to use them and what are they for.

If you want to create a hashtag, simply add # sign followed by the keyword in your twitter update box. For example, you can use something like this:
#wheniambored I play games. If you and others repeatedly use such tags, it will become popular -- or, in other words, it's "Trending" on Twitter. You can also join the conversation by searching hashtags on Twitter search. I hope this helps.
