On the Twitter for iPhone App you go on "Me", then "Followers", then, you click on the person you'd like to unfollow you. On her profile, there'll be a little person with a drop down menu and the option "Block", from there, you just click to block them, and ta-da. To change your settings back I think you have to go on the computer, though. I never tried changing my settings on my phone.
