Can I resubmit a manuscript to a literary agent that has already rejected it, but after modifying it A LOT?

If it's the kind of book that agent sells, then sure. Mention that you've reworked and rewritten it.

Not all agents are alike. If one sells westerns, then don't send him a paranormal!
