Retweet is the way you forward another user's tweet to your followers. While Reply is response to something written by someone on Twitter.

In Retweet There are two options - the Twitter ReTweet without adding your comments, or tweeting with RT with the option to include your comments while in Reply There are two options, a @Reply or DM (direct message reply).

by @username you can send a public message to a person which will be visible in your timeline and the @username messages in the person's profile.
