There are 2 ways of recording with your iSight camera in iMovie.
1. Click the camera button left side middle of your screen.
2. iMovie > File > Import from Camera.
If those 2 don't work then try re-booting your computer and/or doing a file permission repair of your HD in Disk Utility.
