The best way to get Twitter followers is to go to search.twitter.com and enter a keyword which both you and other people have in common. For example if I were into Justin Bieber (which thankfully I'm not)

I'd enter #justin bieber and all the results returned would be people who are likely to follow me. I'd then just add all the users the search found.

I made some software to do this which you can download for free from the resource box below, it will add up to a couple of hundred targeted users per run. You can expect a good percentage of these people to follow you back.
