say i left a message on twitter saying

'hey guys. when is the x factor on tonight?' i no its finished!

and say someone called @anonoymous replied saying '7:30 eeeep i hope joe wins'

how would i know they replied?

You have to click on '@YourUsername' in the main Twitter.com page.

If someone replies to your comment, they will include your twitter tag. ex: if your twitter is @me, their message would be "@me 7:30 eeeep i hope joe wins". Anytime your twitter username is mentioned in someone's tweet, it is called a mention or @reply. You can view these by going to your Twitter home page. On the right, between Home and Direct Messages is @username (in my example, would say @me). Click on that.
