Is there any way to make a tweet unavailable for a certain person to see?

No sorry. Unless they un follow you, then it won't appear on their TL. Yet they can look through your tweets and then see it. Even if you block them, they can still see, sorry.

No, but you can block this person, go to his/her profile, click on the gear icon and select block
