As far as within Twitter, the first to grab it owns it, some celebrities don't own their names on Twitter.

Trademarks are done here USPTO.gov, you don't have to hire an attorney but it might be a bit complicated.

You cannot trademark your twitter user name.

You may however use your trademark name as your twitter username and you don't need to do anything for that apart from using your trademark name as username while registering.Once registered, no one can again use that name. However people may try using the same name with some numbers or letters added to the same and you can not be able to control it but ignore it.

Also if your trademark name is already registered by someone then contact the twitter support. They will help you to release the username and set it free for you to register.
