A lot of trial and error, employing split testing and tracking is essential, there are several stages in the store site purchase process, you should be aware of the percentage of aborted visits at each phase, don't be too scared by high shopping cart abandonment rates, that seems to be a common part of price comparison shopping thee days. Google's free analytics comparable to very expensive services is good for tracking, I believe Google provides split testing support as well, so you can try different sales page variations without looking like you are deceiving the search spiders.

Small time operators have a problem testing small design variations with low traffic. There are more advanced split testing analysis methods offered by some services that can generate meaningful stats with a lower amount of traffic.

There is no such thing as 'perfect' conversion optimisation - it is an ongoing improvement process.

One of the biggest methods is split testing, where multiple version of the same page (or button, or advert etc) are tested with visitors randomly being shown different ones. After a while, you hopefully see one outperforming the others. You can then use this version to create yet another test with more based on the winning version.

Even one you have found the 'perfect' version, competition and markets constantly change, so you will always be looking for the next test or improvement.
