Twitter has multiple uses, but it really depends on what you want to get out of it. It can be used to keep track of the latest news, trends, your favorite celebrities or just a few friends. The '#' is a hash tag and is used to tag key words to help you find what you're looking for. For example if I wanted to see what people are currently saying about a movie that was just released I could search for #MovieTitle and all relevant results would appear.

You will get more followers naturally over time, but if you want people to follow you, then you have to give them a reason to. For your friends and family you'll probably only have to let them know you're using the service. Otherwise you'll have to make your tweets worth reading and find ways to get your twitter username to the masses. Using hash tags (#) and mentioning users directly (@) helps.
