Yes, you can. You can do it by using an @reply. To talk to a person on Twitter you just have to put their username for example @you in the tweet (twitter update) you are tweeting and then tweet it. They will then receive it as a reply. That's how it works. If someone doesn't follow you on Twitter you can't send the person a direct message, it doesn't matter if you follow the person or not.
