If I send someone a direct message on twitter, does it show up on my tweet count?
I've never sent a direct message. If my tweet count is 30, and I send someone a direct message, will my tweet count then be 31?

No.
Direct messages aren't counted as normal tweets, and they aren't visualize in your profile page.
