If I need to add a hashtag, for example #TEST, where do I put it in the tweet? Does it matter if it is in the beginning or does it need to be at the end of the tweet and are you supposed to put the # sign in front of it? Thanks!

Hashtags are a community-driven convention for adding additional context and metadata to your tweets. They're like tags on Flickr, only added inline to your post. You create a hashtag simply by prefixing a word with a hash symbol: #hashtag.
hashtags.org provides real-time tracking of Twitter hashtags. Opt-in by following @hashtags to have your hashtags tracked. It doesn't matter where you put it in your tweet while you need to use # infront of the word. example : #twitter Check out http://twitter.pbworks.com/Hashtags for detailed use and tips.
