It is indeed. Click on Settings in the top navbar. You should automatically be seeing the "Account" tab within the settings. Scroll to the bottom and click on "Protect my tweets" (next to the "Tweet Privacy" section name with a lock icon next to it).

Bam. Now anyone who wants to follow you has to "request" you first, and you get to approve (or decline) these follow requests.
