Start with a websearch using their name and last known location.

Facebook or myspace them, if no luck look on whitepages.com to see if you can find them. Or maybe ask a family member about there where abouts as of now.

Find someone with an Electoral Roll "plumbed" into their computer and ask them to check out your relative (it's only 80% complete but that's often enough to find someone)
If you know the area in which they were living - a letter to the local newspaper - address it to the "Readers' Letters Column" asking if anyone knows what happened to your Aunt Matilda
If you knew the address where they were living but have moved - a letter to neighbours either side may come up with an address. I'm still in touch with people who moved from my street 20 years ago.
Any Clubs and Societies of which they were members - I once found someone I thought was living in the South of England and knew he was interested in Rifle shooting. I wrote to the Rifleman's Society and they passed on my letter to him in Scotland! No wonder I couldn't find him on the Electoral Roll.
