If my twitter account is on private, will people (who are not my followers or I'm not following) that's been mentioned in my tweet be able to read my message?

No, unless they follow you they can't read your tweets even if you write it for them with an @ sign!

If your twitter is on private, hashtags and all the trending topics stuff you tweet don't count to. Sad
