I think if you don't follow any black hat method, 300-500 followers is enough. Be more active, you will get more real followers.

It depends a lot on your on how many you want and if you have a business.
When it comes to personal accounts close friends, family and some people you are interested in is kinnda enough. But if you want to be popular for some reason anything beyond 1000 is good. For business accounts as many followers you got as better since is all about popularity on social networks. |

This is not an easy question to answer. However, I would say about 500 followers. This is because you said the person does not follow back. It would also depend on what the twets were about. Someone who tweeted funny jokes I would expect would have more followers than someone who twisted posts on discussing Latin.
