I think you're wanting to set your account to Private.
Do the following steps -
Go to settings
Edit My account
Select 'Account' from the set of options on the left-hand side.
Go to 'Tweet Privacy' and put a tick mark on 'Protect My Tweets'
That's it.

This way only the people who are already following you, or those you approve of, will be able to read your tweets and see your followers and following lists.
