In the what your doing box you can type in @username and whatever you want to say. Or if you are on a computer, you can hover to the right and when you see an arrow click it and @username will automatically appear and you can say whatever you want.

You just type @[insert twitter name].

Like, if your friend's twitter name is lala1234. In your status update, just type "@lala1234 hey"

When you are on your home page and you see a twitt from someone that you'd like to respond to, move your mouse to the right of that message and click on the arrow. This will put that person's name in your update box and you can type in a reply.

You can also click on the choice direct message (to the right in the center) and move to a new screen. There's a pull down that lets you chose the name of the person you want to send a private message to. No one else will see this.

Here's a youtube video that demonstrates how to get going on Twitter. I hope it helps: http://www.youtube.com/watch?v=OYHUOESHpVk
