iTunes/External Hard Drive Help!?

If I complete this... http://support.apple.com/kb/HT1449 will the new media I import onto my mac automatically be transferred onto my external hard drive? If it does, will it exist on my desktop at all or just on the External Hard Drive? And will I be able to sync my ipod normally, as in will it sync with all the new media I just put in?
